Can anyone explain why I cannot play my FACEDOWN Twister ? I was about to win this duel (I've played lvl40 Marik several times already with this same deck) but this time my Twister won't let me activate it ?????

i don't know.... might be a bug in the new shadow game skill since you would lost at the end phase if you activated twister.

You can't activate Quick Spell cards during the same turn you set them.

No it's the facedown card is a 2nd Twister. One facedown cannot be played. The one in hand is a backup.
