I tried the Unhappy Girl deck on Lvl 40. I've got Unhappy girl on the field and tried to block is Stone Soldier. He used OtC and I tried to negate it with Riryoku Field but it didn't work?!

Are you attacking in the same turn you set your Riryoku Field? And the OTC already set on his field? If that the case, then yes, you can't use Riryoku Field the turn you set it.

It does target the opponent's monster when tributing. Activate it right away.

If you assume that Riryoku Field can't negate OTC, then you're wrong.

Riryoku Field and OtC were already set a round before. So I don't really get it... can Riryoku Field negate OtC like E-Con?

It does negate. You need to activate it when he targets his own monster as the tribute. If you don't, you will miss the timing.
